Antioxidant isoenzyme responses to nickel-induced stress in tobacco cell suspension culture Scientia Agricola
Pompeu,Georgia Bertoni; Gratão,Priscila Lupino; Vitorello,Victor Alexandre; Azevedo,Ricardo Antunes.
Exposure to nickel (Ni) at high concentrations can lead to production of reactive oxygen species (ROS) resulting in oxidative damage at the cellular level. We investigated the antioxidative responses of Nicotiana tabacum cv BY-2 cell suspension to Ni stress (0.075 and 0.75 mM NiCl2) over a 72 h period with special attention to potential alterations in isoenzymes of superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR). Two main SOD isoenzymes were observed, a Mn-SOD (band I) and a Fe-SOD (band II), as well as one CAT isoenzyme and four GR isoenzymes. Activity staining analysis revealed that CAT activity plays a major role in the early response to Ni-induced oxidative stress, particularly when the Ni concentration used was low, whilst a...

Effect of nickel chloride on Arabidopsis genomic DNA and methylation of 18S rDNA Electron. J. Biotechnol.
Li,Zhongai; Chen,Xin; Li,Suoping; Wang,Zicheng.
Background In recent years, nickel (Ni) has been widely applied in industrial and agricultural production and has become a kind of environmental pollution. In this study, the effect of nickel chloride (NiCl2) with different concentrations on Arabidopsis genomic stability and DNA methylation has been demonstrated. The nucleolus variation and 18S rDNA methylation after NiCl2 treatment have been analyzed. Results The results are as follows: (1) The NiCl2 could result in heritable genomic methylation variations. The genomic DNA methylation variations have been detected by methylation-sensitive amplified polymorphism (MSAP) molecular markers, and the result showed that after NiCl2 treatment, there was methylation variation in T0 generation seedlings, and...

Effects of biosorption parameter: kinetics, isotherm and thermodynamics for Ni(II) biosorption from aqueous solution by Circinella sp. Electron. J. Biotechnol.
Alpat,Senol; Alpat,Sibel Kilinç; Çadirci,Bilge Hilal; Özbayrak,Özge; Yasa,Ihsan.
Circinella sp. was employed as a biosorbent for removal of Ni(II) from aqueous solution. The biosorption kinetics and isotherms were investigated. The effect of several parameters, such as biosorbent dosage, contact time, initial concentration, pH and temperature, on biosorption process was evaluated. The kinetic studies indicated that the biosorption followed pseudo-second order kinetic model. Biosorption behaviour of Ni(II) on Circinella sp. was expressed by both Langmuir and Freundlich isotherms. The equilibrium data fit better to the Langmuir model compared to the Freundlich model in concentration range studied (1.0-3.0 mM). The thermodynamic parameters (ΔG0, ΔH0 and ΔS0) were also determined, and it was found that the Ni(II)...

Evaluation of nickel content in Ni-Mg-Al mixed oxides in steam reforming of simulated biogas. Infoteca-e
CORREIA, F. H.; CURADO, I. C.; GHESTI, G. F.; BRAGA, P. R. S.; MARINHO, A. L. A.; MATTOS, L. V.; NORONHA, F. B.; SOARES, I. P.; SCHULTZ, E. L..
Ni-Mg-Al mixed oxides derived from hydrotalcite-like compounds were prepared by a co-precipitation technique, calcined at two temperatures (550 °C and 750 °C) and applied to steam reforming of simulated biogas. Three nickel contents were used in the catalysts (5, 10 and 15%). The catalysts were characterized by X ray fluorescence, N2 sorption techniques, X-ray diffraction and temperature-programmed desorption of CO2. Steam reforming experiments using simulated biogas were carried out at atmospheric pressure and 750 °C, using S/C ratio of 3.2. Catalysts calcined at 750 °C showed nickel and magnesium aluminate phases, resulting in lower density of basic sites. Conversion of CH4 ranged from 97.4 to 99.6% while CO2 conversion ranged from 58.5 to 63.5%. The...

Interactive effects between supplemental ultraviolet-B radiation and heavy metals on the growth and biochemical characteristics of Spinacia oleracea L. Braz. J. Plant Physiol.
Shweta,Mishra; Agrawal,S.B..
The impact of supplemental UV-B (sUV-B) radiation and heavy metals: Cd (68 µmol.kg-1 soil) and Ni (68 µmol.kg-1 soil) singly and in combination was studied on photosynthetic and non-photosynthetic pigments, metabolites, enzyme activities, biomass accumulation and the net primary productivity of spinach (Spinacia oleracea L. var. All Green), a common leafy vegetable plant. Both the stresses individually and in combination resulted in the reduction of photosynthetic pigments, ascorbic acid, and catalase activity, whereas a reverse trend was observed for anthocyanin, flavonoids and proline content, lipid peroxidation and peroxidase activity. The interactive effects of both the stresses were, however, less than additive. Combined treatment of sUV-B + Cd was...

Nickel and vanadium contamination of benthic invertebrates following the "Erika" wreck ArchiMer
Chiffoleau, Jean-francois; Chauvaud, Laurent; Amouroux, David; Barats, Aurélie; Dufour, Aurélie; Pecheyran, Christophe; Roux, Nathalie.
Levels of nickel and vanadium, trace metals found in high concentration in the oil spilled from the "Erika" tanker off Brittany (France) in December 1999 (41 +/- 1 and 87 +/- 3 mg kg(-1) respectively), have been monitored over a one-year period following this accidental event in several benthic invertebrates. Ni and V in mussels (Mytilus edulis) and oysters (Crassostrea gigas) soft tissues were analysed twice a month by graphite furnace atomic absorption spectrometry and a scallop (Pecten maximus) shell daily growth bands were sampled every three bands by laser ablation and analysed by ICP-MS for these two contaminants. Survey data for mollusc tissues were compared with reference data arising from the national monitoring database. Ni concentrations in...

Nickel biosorption by Acinetobacter baumannii and Pseudomonas aeruginosa isolated from industrial wastewater BJM
Rodríguez,Carlos E.; Quesada,Adolfo.
Nickel biosorption ability was evaluated in two bacterial strains: Acinetobacter baumannii UCR-2971 and Pseudomonas aeruginosa UCR-2957, resulting in greatest adsorption at pH 4.5 and a residence time of 100 minutes. Biosorption isotherms showed that the process follows the Langmuir model. The maximum adsorption rates (Nmax) were 8.8 and 5.7 mg·g-1 for A. baumannii and P. aeruginosa, respectively; however, affinity constants suggest that P. aeruginosa (K=1.28) has higher affinity for nickel than A. baumannii (K=0.68). It is suggested that both strains could be used for wastewater treatment, as long as the concentration of Ni2+ is within the range of mg·L-1.

Nickel isotope fractionation during laterite Ni ore smelting and refining: implications for tracing the sources of Ni in smelter-affected soils ArchiMer
Ratie, G.; Quantin, C.; Jouvin, D.; Calmels, D.; Ettler, V.; Sivry, Y.; Cruz Vieira, L.; Ponzevera, Emmanuel; Garnier, J..
Nickel isotope ratios were measured in ores, fly ash, slags and FeNi samples from two metallurgical plants located in the Goiás State, Brazil (Barro Alto, Niquelândia). This allowed investigating the mass-dependent fractionation of Ni isotopes during the Ni-laterite ore smelting and refining. Feeding material exhibits a large range of δ60Ni values (from 0.02 ± 0.10 ‰ to 0.20 ± 0.05 ‰, n=7), explained by the diversity of Ni-bearing phases, and the average of δ60Nifeeding materials was found equal to 0.08 ± 0.08‰ (2SD, n=7). Both δ60Ni values of fly ash (δ60Ni = 0.07 ± 0.07‰, n=10) and final FeNi produced (0.05 ± 0.02 ‰, n=2) were not significantly different from the feeding materials ones. These values are consistent with the very high production yield of...

Although Ni isotopes have been shown to be significantly fractionated in terrestrial samples, their use in continental environmental studies has not yet been evaluated. The present study focuses on an ultramafic (UM) massif (Barro Alto, Goias, Brazil) because such areas are naturally rich in Ni. We present developed lateritic weathering profiles. The goal of the study is to evaluate the potential of using Ni isotopes in environmental continental studies by combining its isotopic signature with mineralogy, in order to better understand the geochemical cycling of Ni in UM settings during weathering. As such, Ni isotope values were measured in samples from the Barro Alto UM complex in the main stages of the lateritic weathering profile of UM rocks, including...

Nickel Isotope Variations in Terrestrial Silicate Rocks and Geological Reference Materials Measured by MC-ICP-MS ArchiMer
Gueguen, Bleuenn; Rouxel, Olivier; Ponzevera, Emmanuel; Bekker, Andrey; Fouquet, Yves.
Although initial studies have demonstrated the applicability of Ni isotopes for cosmochemistry and as a potential biosignature, the Ni isotope composition of terrestrial igneous and sedimentary rocks, and ore deposits remains poorly known. Our contribution is fourfold: (a) to detail an analytical procedure for Ni isotope determination, (b) to determine the Ni isotope composition of various geological reference materials, (c) to assess the isotope composition of the Bulk Silicate Earth relative to the Ni isotope reference material NIST SRM 986 and (d) to report the range of mass-dependent Ni isotope fractionations in magmatic rocks and ore deposits. After purification through a two-stage chromatography procedure, Ni isotope ratios were measured by MC-ICP-MS...

Nickelophilous plants and their significance in phytotechnologies Braz. J. Plant Physiol.
Prasad,Majeti Narasimha Vara.
Nickeliferous soils are invaded predominantly by members of the Brassicaceae, Cyperaceae, Cunoniaceae, Caryophyllaceae, Fabaceae, Flacourtiaceae, Euphorbiaceous, Lamiaceae, Poaceae and Violaceae, and many of these plants are metal tolerant. About 300 Ni hyperaccumulating plants been identified. These members exhibit unusual appetite for toxic metals and elemental defense. Hyperaccumulators provide protection against fungal and insect attack. Investigations suggested that Ni-hyperaccumulation has a protective function against fungal and bacterial pathogens in Streptanthus polygaloides and Thlaspi montanum. Significance of nickelophilous plants and their significance in phytotechnologies are discussed in this paper.

Response of Crotalaria juncea to nickel exposure Braz. J. Plant Physiol.
Cardoso,Patrícia Felippe; Gratão,Priscila Lupino; Gomes-Junior,Rui Alberto; Medici,Leonardo Oliveira; Azevedo,Ricardo Antunes.
The response of Crotalaria juncea seedlings to nickel (Ni) was investigated. Ni was shown to accumulate mainly in the root system, with little being translocated to the shoots. Catalase (CAT) and glutathione reductase (GR) responses to Ni were also analyzed. CAT activity did not exhibit a clear trend in response to Ni exposure, whereas GR activity appeared to respond to the stress induced by Ni. The results suggest that in C. juncea GR participates in the detoxification of Ni-induced reactive oxygen species via the glutathione-ascorbate cycle.

Solubility and availability of micronutrients extracted from silicate agrominerals PAB
Marchi,Giuliano; Guelfi-Silva,Douglas Ramos; Malaquias,Juaci Vitória; Guilherme,Luiz Roberto Guimarães; Spehar,Carlos Roberto; Martins,Eder de Souza.
Abstract: The objective of this work was to evaluate the solubility of the micronutrients copper, nickel, and zinc, chemically extracted from silicate agrominerals, as alternative sources of potassium for lettuce (Lactuca sativa) and rice (Oryza sativa) cultures, as well as their availability to the plants. The micronutrient contents of five agrominerals (alkaline ultramafic, biotite schist, volcanic breccia, phlogopite schist, and mining by-product from Chapada) were evaluated, after being extracted by the citric acid, hydrochloric acid, diethylenetriaminepentaacetic acid, 3051A, neutral ammonium citrate, and Mehlich-3 methods. The agrominerals, at the doses of 0 (control), 200, 400, and 600 kg ha-1 K, were mixed to soil where lettuce and rice were...

The behavior of nickel isotopes at the biogeochemical interface between ultramafic soils and Ni accumulator species ArchiMer
Ratie, G.; Quantin, C.; Maia De Freitas, A.; Echevarria, G.; Ponzevera, Emmanuel; Garnier, J..
Ultramafic derived soils are characterized by low nutrient soils, a low Ca:Mg ratio, and high metal contents such as Ni, Co and Cr. The vegetation growing on these soils is highly adapted and includes both Ni hyperaccumulator and accumulator species. Today, approximately 530 Ni hyperaccumulator species are listed worldwide and the Ni concentration can be extremely high, e.g. up to 25% in latex from Pycnandra acuminata (Sapotaceae), a tree found in New Caledonia. The aim of this study is to identify the potential role of Ni hyperaccumulator plants in the Ni biogeochemical cycle at the soil surface by using Ni isotopes. A set of Ni hyperaccumulator and Ni accumulator plants as well as topsoils were sampled on the Barro Alto and Niquelândia ultramafic...

Trace element systematics in cold seep carbonates and associated lipid compounds ArchiMer
Wang, Xudong; Bayon, Germain; Kim, Jung-hyun; Lee, Dong-hun; Kim, Dahae; Guéguen, Bleuenn; Rouget, Marie-laure; Barrat, Jean-alix; Toffin, Laurent; Feng, Dong.
Seeping of methane-rich fluids at submarine cold seeps drives intense microbial activity and precipitation of authigenic carbonates. Some trace elements play an important role in the biogeochemical processes operating at cold seeps, especially as specific enzymatic co-factors related to methanogenesis and the anaerobic oxidation of methane (AOM). However, it is unclear whether microbial trace metal utilization can be traced by the geochemical composition of seep carbonates. In this study, we analyzed a series of authigenic carbonate samples recovered from various seep settings worldwide and report for the first time trace element concentrations for total lipid fractions, combined with biomarker analyses and determination of elemental abundances in...
